(Finding Bacteria in School)
Purpose
To find out where most bacteria are in school. To analyze what we find and Gram Stain it to find out if it is Gram Positive or GramNegative.
Hypothesis
If the doorknob is swabbed, then the most amount of bacteria will be seen.
Materials
* 1 Q-tip
* 1 Permanent Marker
* 1 Petri dish with nutrient agar
* BunsenBurner
* Inoculating Loop
* Gram Stains
Safety Precautions
We will be using a Bunsen burner so we will need to be careful with the open flame. We will also need to clean everything before andafter use because we are working with bacteria. And because of both the bacteria and the flame, we will need to wear goggles, and not wear open-toed shoes, dangling jewelry, and we will need to keeplong hair tied back.
Procedure
A partner was found. A petri dish was divided in half and names and periods were written in each half. At each specific location, a Q-tip was used to rub the surfacebeing tested. The Q-tip was then gently rubbed onto the agar surface, taking care not to tear or dig the agar. A hypothesis was then written. After that, the petri dish was put in the incubator and leftfor 2 days. After 2 days, goggles were put on, the lab table was cleaned with disinfectant, and the petri dish was found in the incubator. After taking it out, the results were noted in the datatable and the colonies compared and the lid was kept on the whole time. A slide was cleaned with disinfectant, rinsed with water, and dried using a cloth. A very small amount of distilled water was placedon the slide using a pipette. The inoculating loop was sterilized by passing it through the flame of the Bunsen burner until the end turned red-hot. After it cooled, a very small amount of culturefrom the agar surface was removed and it was touched to the drop of water until it turned cloudy.
