Como como cadena individual de ADN , cada nucleotido induce cambios en una corriente electrica que peude se traducidaen la secuencia de datos.
Un beneficio clave de esta plataforma esque puede obtener lecturas mucho mas largas que los tipicas 100-200 pares de bases que por lo general se obtienen con lasplataformas existentes.
El sistema puede tomar una secuencai de ADN directamente de la sangre.
Las maquians que leen ADN y ARN siguen costando cientos de miles de dolares , ocupan mucho espacio y requierenmucho mas proceso inicial y postsencuenciacion, para generar un genoma.
The theory behind nanopore sequencing is that when a nanopore is immersed in a conducting fluid and a potential (voltage) isapplied across it, anelectric current due to conduction of ions through the nanopore can be observed. The amount of current is very sensitive to the size and shape of the nanopore. If singlenucleotides (bases), strands of DNA or other molecules pass through or near the nanopore, this can create a characteristic change in the magnitude of the current through the nanopore.
Alpha hemolysin(αHL), a nanopore from bacteria that causes lysis of red blood cells, has been studied for over 15 years.[5] To this point, studies have shown that all four bases can be identified using ionic currentmeasured across the αHL pore.[6][7] The structure of αHL is advantageous to identify specific bases moving through the pore. The αHL pore is ~10 nm long, with two distinct 5 nm sections. The uppersection consists of a larger, vestibule-like structure and the lower section consists of three possible recognition sites (R1, R2, R3), and is able to discriminate between each base.[6][7]
