X-4 Samples produced
X-4.1 Platelet rich plasma (PRP) prepared :
After addition and mixing of sodium citrate to the whole blood, centrifuge at 500-800 rpm, 25 T? -32 “C,for 10 minutes. After the centrifuge stops, transfer the PRP into a test tube with a pipette.
☆ Attention:
a. Ideal ratio of PRP: whole blood = 1 : 4. If PRP is less than this ratio, the amount ofPRP can be increased by extending the centrifugation time, while keeping the rotation speed constant. If the PRP ratio is too high, then re-centrifuge with either a reduced rotation speed, or for ashorter period of time.
b. Under excessive centrifugal forces, platelet specimens will sink, especially larger plaque, and the plaque aggregation response will be stronger.

d. When transferring PRP, take care not to contaminate with RBCs. Contamination can be seen by the naked eye, in the form of small particles.This is to ensure the reliability of the samples.
X-4.2 Platelet poor plasma (PPP) prepared :
Once the PRP has been drawn to the blood centrifuge, centrifuge at 2000-2500 Turn/Min for about 10minutes, for PPP.
Attention: Normal PPP and PRP prepared by the naked eye to, as there are distinct differences between the two. For example, PPP is brighter than PRP.
X-4.3 For platelet gatheredexperiments, the platelet count should be taken. Ideal PRP samples will have a platelet count of (200 x 10A9)/L ~ (250 x 10A9) /L. If the platelet count is less than (100 x 109) /L or larger than (1000 x 10A9)/L, the platelet accumulation response will be lower.
X-5 Samples available
X-5.1 After PRP is gathered wait 30 minutes before measuring platelet accumulation, as platelet reaction is poor.However, the measurement must be completed within 3 hours.
X-5.2 Collected blood samples should be stored between 15-25^0. Cold environments may induce spontaneous aggregation of the sample.
