DNA from Kiwi

Material per group:

50ml alcohol, 800ml beaker, kiwi, knife, 400ml beaker, mortar and pestle, balance, 100 mldigestion solution (1.25 g tri-sodium citrate dihydrate = citric acid, 3g salt, 1 drop of dish detergent), 10ml dish detergent, thermometer, clock, ice cubes, 50ml ice cold alcohol, funnels, filters,small glass, wood skewers

Procedure:
* Put 50ml alcohol in freezer
* Heat water bath to 60 °C
* Peel kiwi, homogenize it in mortar coarsely
* Mix homogenized kiwi in 400ml beaker with100 ml of digestion solution
* Ad10ml dish detergent to kiwi, swirl gently
* Put kiwi 15-20 minutes in hot water bath
* Prepare bowl with ice cubes and cold water
* Put kiwi 5minutes in ice bath
* Filtrate kiwi mix
* Overlay with extreme caution the kiwi filtrate with 50 ml ice cold alcohol
* Wrap DNA with wooden skewer, store it in alcohol

Research Question: How to extract DNA from kiwi cells?

Conclusion:

In this experiment we followed a number of steps in order to extract DNA from a kiwi;our objective was to prove that all living organisms are made of cells which contain DNA in their nucleus.

The first step was to peel a kiwi and to mash it in a mortar until we got a homogenizemixture; we did so in order to break the cell walls and cause cell disruption. Our next step was to mix our homogenize mixture with 100ml of a digested solution which contained 3g of salt, 1.25g of acitric solution, 10g of dish detergent and water, water allows the uniformity of the reactions.
