Studies of aloe. VI. Cathartic effect of isobarbaloin.

Abstract
The cathartic effect of isobarbaloin, a stereoisomer of barbaloin (compound principally responsible for the cathartic activity of Aloe), was examined in male rats by oral administration. Individual differences insensitivity in the laxative activity of isobarbaloin and barbaloin was not found. The cathartic activity (ED50) of isobarbaloin in barbaloin positive rats was 19.2 mg/kg, nearly equal to that of barbaloin (19.5 mg/kg). Also, isobarbaloin administered orally was demonstrated to decompose to aloe-emodin-9-anthrone (active metabolite of barbaloin) as well as to barbaloin. Therefore, it is considered that themechanism underlying the cathartic effect of isobarbaloin is the same as that of barbaloin.

Abstract
Methods using liquidchromatography/mass spectrometry (LC/MS) and LC with diode array detection (DAD) in the UV range (LC/UV) were developed for the determination of low levels of the anthraquinones aloe-emodin and aloin-A (barbaloin) in aloe-based products. The methods were used to analyze several commercial products (liquids, semisolids, and solids) for the 2 anthraquinones. The wavelengths used for quantification ofaloin-A, aloe-emodin, and emodin (internal standard) by DAD were 357, 257, and 289 nm, respectively. The on-column sensitivities were 0.25 and 0.05 ng by LC/UV and 0.01 and 0.025 ng by LC/MS for aloin-A and aloe-emodin, respectively. The methods are simple and sensitive and provide reproducible results; therefore, they are suitable for the determination of these anthraquinones in various aloe-basedproducts.

Abstract
Aloe-emodin-9-anthrone(AE-anthrone), produced from barbaloin in the rat large intestine, caused not only an increase in the intestinal water content but also stimulated mucus secretion. This might play an important role inthe occurrence of diarrhea. It was demonstrated that the amount of AE-anthrone produced in the rat large intestine(maximal amount: 568 micrograms/rat at 4 h after injection) was enough to cause both of these effects, which were observed following intracecal administration of barbaloin (31.1 mg/kg). These results together with our previous data, which showed a relationship between increase in theintestinal water content and the stimulation of peristalsis, confirm that AE-anthrone is the principal agent responsible for the cathartic effect of barbaloin. We also propose that the increase in water content is a more important factor than stimulation of peristalsis in the induction of diarrhea by barbaloin.

En el marco del desarrollo de la cadena nacional dela sábila, se han suscitado varias preguntas de tipo técnico que seguramente en otros países mas adelantados en el tema se han resuelto con éxito, esto debido a que en el tema de la investigación estamos retrazados mas de treinta años, esto sería normal y silvestre si el retrazo fuera con respecto a las grandes potencias como EEUU y la comunidad económica europea (CEE), pero además de estosestamos atrasados con respecto a países centro y suramericanos como: México, Venezuela y Argentina. En donde el desarrollo de este tipo de cultivos y el aprovechamiento de los mismos ha venido ligado de la mano de la investigación en laboratorios y aulas, obviamente con el apoyo de sus gobiernos y de los industriales, que en últimas son los más beneficiados.
