DNA EXTRACTION


Objective: to see if DNA looks the same with different organisms (apple and banana)


Introduction: Deoxyribonucleic acid (DNA) is a nucleic acid that contains the geneticinstructions used in the development and functioning of all known living organisms and some viruses. The main role of DNA molecules is the long-term storage of information. DNA is often compared to a setof blueprints or a recipe, or a code, since it contains the instructions needed to construct other components of cells, such as proteins and RNA molecules. The DNA segments that carry this geneticinformation are called genes, but other DNA sequences have structural purposes, or are involved in regulating the use of this genetic information.

Hypothesis: when we take out DNA from differentorganisms, then we will see that the structure of the DNA is different in each one of them.

Materials:
- Knife
- Apple
- Banana
- Cheesecloth
- Mortar
- Small funnel
-Ethanol
- Graduated cylinders
- 7 ml DNA buffer
- Glass stirring rod
- Blender

Methods:
a) A piece of banana was smashed with a mortar.
b) 7 ml of buffer were added to the banana puree.
c) Thefunnel was put over the graduated cylinder and the cheesecloth was positioned on top of the funnel.
d) The smashed banana was drained on top of the cheesecloth into the graduated cylinders.
e) Anequal amount of alcohol compared to the banana juice was poured in the graduated cylinder.
f) Using the stirring rod, DNA was collected at the boundary of alcohol and banana juice and its appearance wasexamined.
g) An apple was cut with a knife and then smashed with a blender
h) 7 ml of buffer were added to the smashed apple
i) The funnel was put over the graduated cylinder and the cheeseclothwas positioned on top of the funnel.
j) The apple was drained on top of the cheesecloth into the graduated cylinders.
